I would like to know how to reach Kassandra (Afitos) from thessaloniki airport by bus.

Is it possible or do i have to go to thessaloniki centre before?

And once i'm in Kassandra can i move in it by local bus or it's necesary to rent a car/motorcycle?

As far as I know, you can not take the bus from the airport. You have to go to the terminal first by taxi or city bus.
